Obviously, the easiest way to do this is to just clear the first two floors carefully with either Isaac or Maggy as they have decent starting HP which will keep you alive without needing to worry too much. This includes both soul and demon hearts. To unlock Eve you must clear two floors in a row without picking up any heart items.
